Abstract:
Meiosis is an important generator of genetic diversity in plants. Maize, with its large and conspicuous chromosomes, is an attractive model for cytological studies of meiosis. Immunolocalization employs the antigen-antibody interaction to visualize proteins within cells and tissues using fluorescence microscopy. This technique enables direct observation of protein distribution and dynamics in the native chromosomal context, revealing spatial and temporal patterns of nuclear and cellular processes. This protocol describes the procedure to immunolocalize proteins in meiotic cells of maize. To conduct immunolocalization, anthers at appropriate meiotic stages are dissected and fixed. Meiotic cells are released from anthers, affixed to slides, and incubated with primary antibodies, followed by incubation with fluorescently labeled secondary antibodies. Fluorescent signals are examined with fluorescence microscopy. In this way, the location of proteins participating in meiosis can be identified. Protein colocalization can also be determined. Finally, this method can be easily adapted to use to examine protein localization in somatic cells.
