Incubation and physiological readouts in vitro for thiamine triphosphate and related compounds in plants

Abstract:
Thiamine triphosphate (ThTP) is a low-abundance thiamine derivative the metabolism of which in plants remains poorly understood because of its dynamic behavior, apparent lability, and strong dependence on subcellular context. In this chapter, we describe an integrated experimental framework for investigating ThTP and related thiamine derivatives in plants. The workflow combines isolation and incubation in vitro of chloroplasts, HPLC-based quantification of thiamine derivatives, enzymatic verification of ThTP peak identity, and electrochromic shift (ECS) measurements as a physiological readout of altered ThTP levels. Together, these methods provide a practical platform for examining the metabolic significance of this minor thiamine derivative in plant systems.
