Abstract:
Bacillus sp. HMM can efficiently yield cellulase. However, transcriptome data and sequences of important genes such as cellulase gene are indistinct now, which restricts genetic reconstruction of strain HMM. In current paper, GO, COG and KEGG classification of unigenes from transcriptome sequencing of strain HMM was analyzed in detail, and cellulase gene GH (TR1834) encoding glycosylhydrolase was selected. Moreover, transgenic Bacillus sp. HMM overexpressing GH (GHT) with recombinant plasmid PHY-300PLK-GH was constructed. Avicel activity, CMC-Na activity and filter paper activity of GH transgenic HMM (GHT) were 1.4-, 1.2- and 1.08-fold those of control Bacillus sp. HMM with empty plasmid PHY-300PLK. This study enhanced application value of Bacillus for hydrolysis of cellulose.
