Abstract:
Flow cytometry has made it possible to identify human B cell subsets based on surface and intracellular antigens, a process known as immunophenotyping. CD19 and CD20 are commonly expressed in all B cells; the presence or absence of CD27 separates memory B cells from naïve cells, while IgD and IgM are used to distinguish naïve from transitional B cells. Markers such as CD38, CD27, and CD24 are helpful to identify plasmablasts and immature B cells. CD21 points to mature B cells, CD10 is on immature B cells, CD138 marks plasma cells, and CD5 is high in regulatory B cells. Memory B cells have less CD23 than naïve B cells. B cells are multifaceted. They present antigens (APCs), secrete antibodies, and keep the immune system in check. Identifying B-cell subsets in clinical settings-especially for diagnosing immune disorders, autoimmune diseases, and blood cancers like leukemias and lymphomas. Knowing which B cells are present in a certain disease setting helps clinicians understand how far a disease has progressed and how active it is. With this information, a treatment that fits the patient's needs can be selected, ones that target B cells directly, like anti-CD20 antibodies. With flow cytometry as a tool, we can spot B-cell malignancies like chronic lymphocytic leukemia (CLL), follicular lymphoma, mantle cell lymphoma, and multiple myeloma. Flow cytometry is a great tool for both clinicians and researchers. It is fast, reliable, and sensitive. In our protocol, we used a 19-fluorochrome panel to successfully sort out different B cell subsets from human blood. Advances like these not only help classify the disease better, but they also make it possible to personalize treatment decisions in ways that matter for the patients.
