Arrayed Magnetic Accelerator Synergistically Enhances Orthodontic Tooth Movement via Microtubule Assembly-Mediated

Abstract:
Mechanical orthodontic treatment is regarded as a classical strategy for directional tooth movement in confront of malocclusion. However, prolonged remodeling period and adverse complication risks, which are mainly ascribed to inadequate sensation of bio-mechanical cues, predominantly restrict orthodontic clinical outcomes. This study demonstrates that construction of biophysical static magnetic fields (SMFs) is highly involved in accelerated tooth movement and bone remodeling in manners dependent on amplification of mechanical sensing. The arrayed magnetic orthodontic accelerator (AMOA) with precisely controllable SMFs was fabricated by 3D printing. Exposure to SMFs can enhance production of pro-inflammatory cytokines from periodontal ligament cells (PDLCs) through mechanical sensitization to facilitate polarization of M1-like macrophages and differentiation of osteoclasts for improved bone remodeling. Pre-clinical exploitation clarifies that AMOA application dramatically facilitated alveolar bone remodeling and osteoclastic activity in the compression side of orthodontic tooth model. In summary, our results suggest that SMFs-upregulated sensing of biological force in PDLCs participates in orthodontic tissue remodeling and plays a pivotal role in acceleration of tooth movement. This study sheds light on the therapeutic transformation of AMOA to improve clinical outcomes of classical orthodontic treatment.
