Abstract:
Objective: Jujuboside A (JuA) possesses anti-inflammatory and antioxidant activities; however, its role and mechanisms of action in ischemic stroke (IS) remain unclear.Methods: A mouse MCAO was established to evaluate the effects of different doses of JuA on brain water content, neurological function scores, behavioral parameters, inflammatory factors, and oxidative stress markers. An OGD model was established in SH-SY5Y cells. Cell viability and apoptosis were assessed by the CCK-8 assay and flow cytometry. Inflammatory factors were evaluated via ELISA, and oxidative stress markers were assessed by commercial kits. RT-qPCR was used to analyze changes in miR-583 expression under OGD conditions.Results: JuA dose-dependently alleviated cerebral edema, neurological deficits, and motor dysfunction in MCAO mice; it reduced TNF-α, IL-6, IL-1β, MDA, and LDH, and increased SOD levels. Under OGD conditions, JuA (40 μM) significantly improved SH-SY5Y cell viability and reduced cell apoptosis, inflammatory responses, and oxidative stress. Furthermore, inhibition of miR-583 abolished the protective effects of JuA.Conclusion: By upregulating miR-583 to suppress inflammation and oxidative stress, JuA alleviates IS injury and represents a promising candidate neuroprotective compound.
