Abstract
Background:
We evaluated the safety and efficacy of ponatinib, a multi-target tyrosine kinase inhibitor, for pulmonary tumor thrombotic microangiopathy (PTTM).
Methods And Results:
We conducted a single-center, open-label, single-arm, prospective study; 3 patients associated with ovarian, breast, and gastric carcinomas were enrolled and received oral ponatinib for 2 weeks. All died from PTTM progression with no severe ponatinib-related adverse events.
Conclusions:
This study failed to demonstrate ponatinib efficacy. Further studies may evaluate the effect of different biochemical targets on PTTM.
