Abstract:
Epidemiological studies have established obesity as a risk factor for multiple cancer types. Tumor angiogenesis, an indispensable process sustaining tumor growth and metastasis, serves as a critical bridge between obesity and malignancy. However, how the diverse components of the adipose microenvironment coordinate to stimulate tumor cells to support angiogenesis remains poorly understood. This review highlights the central role of adipose tissue as a key regulator of tumor angiogenesis, discussing how its secreted factors and cellular constituents collectively promote angiogenesis in the tumor microenvironment. It also examines the current status of clinical translation for adipose-targeted anti-angiogenic strategies, summarizing early progress and existing gaps. By elucidating adipose tissue-vascular crosstalk, this review aims to provide a foundation for developing novel anti-angiogenic therapies for obesity-associated cancers.
