Abstract:
This chapter details the methodology for generating DNA barcoded strains of Salmonella using a tri-parental conjugation. While this protocol was developed using S. typhimurium, it should remain applicable to additional Salmonella serotypes as well as other Gram-negative bacteria. The subsequently constructed library of DNA barcoded strains can be implemented, when complemented with next-generation sequencing or quantitative PCR, to quantitatively examine the impact of this pathogen on bacterial populations or transmission throughout a host.
