Abstract:
Allergic diseases affect approximately 22% of the global population. Rapid and accurate identification of causative allergens is essential for enabling timely clinical intervention and preventing re-exposure. Current clinical approaches for in vivo allergen diagnosis often involve risks associated with direct allergen exposure, while many existing in vitro diagnostic methods remain labor-intensive, time-consuming, and technically demanding. For example, the basophil activation test requires sophisticated instrumentation and specialized operational expertise. Therefore, there is an urgent need for a convenient, rapid, sensitive, and easily assembled platform for reliable in vitro allergen diagnosis while reducing reliance on sample pretreatment, auxiliary instrumentation, and external control systems. In this study, a disc-shaped microfluidic device, termed the relative rotation SlipChip (rr-SlipChip), was developed. Unlike conventional SlipChip platforms that depend on precise microstructural alignment, the rr-SlipChip employs radial alignment and step-by-step rotation to enable convenient configuration of loading pathways for multiple immunoreagents. In addition, the reagent pathways are completely isolated from one another, thereby minimizing the risk of cross-contamination. Using a single rotary pneumatic control channel combined with a simple sliding operation, the proposed device enables immunoassays to be completed within approximately 25-37 min. The rr-SlipChip achieves favorable analytical sensitivity, with limits of detection of 0.614 ng mL-1 for sIgG and 0.689 ng mL-1 for sIgE. Therefore, the rr-SlipChip introduces a new rotational sliding microfluidic architecture distinct from conventional SlipChip designs, providing a novel platform for rapid, sensitive, continuous, and scalable in vitro allergen diagnosis with significant potential for point-of-care testing (POCT) and accessible precision allergy management.
