Abstract:
BruUV-seq is a nascent RNA sequencing technique that uses bromouridine to label RNA and UV light to suppress both transcription elongation and RNA degradation, enabling the enrichment and study of labile RNA species, such as enhancer RNAs (eRNAs), in live-cell culture models. Here, we describe both the BruUV-seq experimental method and the next-generation sequencing data analysis, which allow for the identification of eRNAs produced at active enhancer elements.
