Abstract:
MicroRNA-570 (miR-570), particularly miR-570-3p, has emerged as a context-dependent regulator of cancer biology with both tumor-suppressive and oncogenic functions. This review summarizes current evidence on the molecular regulation, functional roles, genetic variation, biomarker potential, and therapeutic targeting of the miR-570 network. miR-570 expression is controlled by epigenetic remodeling, DNA methylation, genomic copy-number variation, competing endogenous RNAs, and extracellular-vesicle transport. In tumor-suppressive settings, miR-570 restrains proliferation, metabolism, proteostasis, invasion, immune-checkpoint expression, stemness, treatment resistance, and metastasis through targets including DPP4, PD-L1, SPP1, IRS1/2, LCMR1, ATG12, and chaperome components. Conversely, it may promote malignancy by repressing MX2, FOXO3, PTEN, KLF9, and KLF10. These opposing effects are shaped by tumor lineage, mature strand, cellular source, target abundance, ceRNA buffering, genetic variants, and treatment state. Clinical studies suggest context-dependent diagnostic and prognostic relevance, while evidence for predictive or pharmacodynamic applications remains insufficient due to methodological heterogeneity, limited external validation, and inconsistent strand-specific measurement. Therapeutic restoration, inhibition, ceRNA targeting, and epigenetic reactivation remain promising but require molecular patient selection and precise cell-specific delivery. Future translation depends on standardized assays, direct target validation, cell-resolved profiling, and rigorous preclinical safety assessment. Accordingly, miR-570 should be viewed as a context-defined network rather than an isolated biomarker.
