Abstract:
Liu et al. enabled genome-wide clustered regularly interspaced short palindromic repeats (CRISPR) screening of primary human T cells in vivo and identified P2RY8 and GNAS through complementary functional readouts. We discuss how context shapes perturbation effects, and how readouts guide prioritization, with implications for T cell engineering in solid tumors.
