Abstract:
The network of transcription factors (TFs) is an integral component of executing macroautophagy/autophagy-lysosome gene expression to maintain cellular homeostasis. Across metazoans, conserved TFs, particularly the helix-loop-helix factor TFEB, convert upstream signals into specific genetic programs through mechanisms ranging from DNA recognition and chromatin remodeling to biomolecular condensates. The dysregulation of TF-governed gene programming disrupts the progression of metabolic disease and cancer, highlighting that dissection of transcriptional regulation makes it a promising therapeutic target. Consequently, instead of indirectly modulating the pathways in which they are involved, targeting these essential TFs directly is becoming a new trend for pharmaceutical development. In this review, we systematically elucidate current mechanisms of the TF-driven autophagic transcriptome, and summarize current small molecules targeting these TFs in various disease.
