Abstract:
Parkinson's disease is a common, highly prevalent and severely debilitating progressive neurodegenerative disorder. However, despite extensive clinical and preclinical research, the pathobiological complexity, underlying mechanisms and treatment strategies of this disorder remain poorly understood. Animal models are a valuable tool to study human brain disorders, including Parkinson's disease. Here, we discuss the pathobiology of Parkinson's disease, with a particular focus on zebrafish (Danio rerio) based genetic, pharmacological, and behavioral models that provide important insights into genetic and environmental causes of this disorder. Critically evaluating the advantages and limitations of modern zebrafish models relevant to Parkinson's disease, we emphasize the importance of addressing both motor and nonmotor symptoms of this disorder, and also outline how these fish can facilitate further anti-Parkinson's disease drug discovery.
