Abstract:
Hydrogel coating is one of the most promising solutions to address a series of issues caused by the mechanical mismatch between implantable medical devices (IMDs) and human tissues. However, existing methods to construct hydrogel coatings on IMDs typically require harsh preparation conditions. In this work, a trigger-free interfacial click chemistry-protein chemical coupling (IC-PC) strategy is proposed to build hydrogel coating on IMDs surface. By using BSA as a protein model, a BSA hydrogel coating with strong bonding (adhesion strength of 200-550 kPa) was successfully prepared on diverse IMDs via disulfide bond/sulfhydryl click chemistry and EDC/NHS coupling chemistry. Through fluorescence experiments, the adhesive mechanism was concluded to be the combination of a rapid interfacial click chemistry and a slow EDC/NHS-mediated coupling. Moreover, a bioactive luteolin (Lut) was loaded into the BSA hydrogel coating, and the obtained Lut@BSA hydrogel coating presented prominent bio-properties, including antifouling, ROS scavenging, and hemocompatibility. Finally, the ex vivo blood circulation experiment indicated that no thrombus formed on the Lut@BSA hydrogel coating, which verified its application potential for blood-contacting catheter. We believe this strategy will provide insights into novel design methods of hydrogel coating construction.
