Abstract:
Airway hyperresponsiveness and remodeling are major features of asthma. We investigated whether pharmacological activation of the aryl hydrocarbon receptor (AhR) modifies these responses in a mixed allergen (MA) mouse model of asthma. MA exposure increased airway resistance, contractility, inflammation, and remodeling, with elevated AhR expression in whole lung and airway smooth muscle but limited canonical AhR target gene activation. Treatment with the high-affinity AhR agonist 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) improved lung mechanics, reduced inflammatory cell and cytokine accumulation, attenuated airway remodeling, and decreased airway narrowing in precision-cut lung slices. Bulk lung RNA sequencing further showed suppression of inflammatory, neutrophil-associated, and remodeling pathways following TCDD treatment, together with induction of canonical AhR target genes. Collectively, these findings demonstrate that AhR activation suppresses inflammatory gene networks, limits airway remodeling, and improves airway mechanics in experimental asthma, supporting the therapeutic potential of targeting AhR signaling to mitigate airway dysfunction and structural remodeling.

Asthma: Pathogenesis and Management
Asthma is classified as allergic and non-allergic. Allergens such as dust mites, pollen, and pet dander trigger allergic asthma, while factors like cold air, intense emotions, or exercise can induce non-allergic asthma.

Antiasthma Drugs: Mast Cell Stabilizers and Anti-IgE Drugs
Mast cell stabilizers, such as cromolyn (also known as sodium cromoglycate) and nedocromil (Tilade), are effective drugs in asthma management. These stabilizers hinder histamine release by skillfully obstructing the activation of mast cells and other cellular entities. Notably, they navigate this task without...
Antiasthma Drugs: β2-Adrenoceptor Agonists
One class of bronchodilators includes β2-adrenoceptor agonists. These agents target the β2-adrenoceptors located on bronchial smooth muscle cells. By stimulating these receptors, β2-agonists induce relaxation in these...
Antiasthma Drugs: Muscarinic Receptor Antagonists
Antimuscarinic agents compete with ACh for the same binding site on the muscarinic receptors. By binding to these receptors, they inhibit the downstream effects of ACh and block the parasympathetic...
