Abstract:
Stoichiometry is often treated as a decisive test of post-translational modification significance, but low occupancy does not necessarily imply low biological importance. Cysteine oxidation exposes this limitation: distinct sulfur states differ in lifetime, reversibility, repairability, and function. Recent redox-proteomic methods quantify oxidation alongside protein abundance and resolve localization and cell-state context, yet occupancy, exposure, and flux remain distinct quantities. Here, I review quantitative examples from cysteine redox biology and other protein modifications, arguing that stoichiometry is a mechanistic constraint rather than a verdict. Low modified fractions can be consequential through catalytic amplification, temporal control, or local enrichment, but these mechanisms require functional evidence. Irreversible probes reveal transiently populated states through cumulative capture, providing useful comparisons under matched conditions rather than measuring instantaneous occupancy or native redox cycles. The central questions are what chemical state is measured, in which molecular population, over what time window, and with what functional leverage.
