Introduction:
Osteoarthritis (OA) remains a degenerative joint disorder with limited treatment options.
Methods:
We examined miR-302c-3p expression in human OA cartilage and primary chondrocytes, validated ADAMTS5 as a direct target, and developed a PCEC/TAmP hydrogel for intra-articular delivery in a DMM mouse model.
Results:
miR-302c-3p was downregulated in OA cartilage and attenuated IL-1β-induced chondrocyte catabolism by reducing ADAMTS5, MMP1, and MMP13 expression while preserving COL II. The PCEC/TAmP hydrogel enabled sustained release and improved cartilage penetration of miR-302c-3p, enhancing its therapeutic efficacy in OA mice.
Conclusion:
miR-302c-3p-loaded PCEC/TAmP hydrogel may be a promising intra-articular strategy for targeted OA treatment.
