Abstract:
Human induced pluripotent stem cells (hiPSCs) are a valuable system for studying human development, modeling disease-associated cellular phenotypes, and generating difficult-to-access cell types, such as sensory neurons (SNs). Here, we present a stepwise differentiation protocol for generating peripheral SNs from hiPSCs under defined culture conditions. We describe steps for expansion of hiPSCs, induction of neural crest cells (NCCs), dissociation and replating of NCCs, and maturation into SNs. This protocol yields nociceptor-enriched SN cultures.
