    Summary

    This study found no evidence of dopamine hypersensitivity in the tuberoinfundibular dopamine (TIDA) system in patients with tardive dyskinesia. Prolactin responses were similar to control groups, suggesting this pathway is not affected.

    Background:

    • Tardive dyskinesia is linked to dopamine hypersensitivity in the nigrostriatal system.
    • The tuberoinfundibular dopamine (TIDA) system regulates prolactin secretion via inhibition.

    Purpose of the Study:

    • To investigate if dopamine hypersensitivity extends to the TIDA system in tardive dyskinesia patients.
    • To assess pituitary prolactin secretion in response to dopamine modulation.

    Main Methods:

    • Assessed plasma prolactin in six tardive dyskinesia patients under four conditions.
    • Conditions included chronic haloperidol, withdrawal, unmediated state, and acute haloperidol dose.
    • Compared prolactin responses with normal subjects and schizophrenic patients without tardive dyskinesia.

    Main Results:

    • No significant differences in prolactin responses were observed across the four conditions.
    • Prolactin responses in tardive dyskinesia patients did not differ from control groups.
    • This indicates no post-synaptic dopamine hypersensitivity in the TIDA-pituitary pathway.

    Conclusions:

    • The study provides no evidence for TIDA-pituitary pathway dopamine hypersensitivity in tardive dyskinesia.
    • Neuroleptic-induced dopamine hypersensitivity may not occur in the human TIDA-pituitary system.
    • Findings are consistent with previous studies on hormonal responses to dopamine agonists.
