    Summary

    New adipocyte synthesis in Sprague-Dawley rats occurs postnatally and ceases before sexual maturity. A significant population of precursor cells (preadipocytes) is generated during the early postnatal weeks.

    Background:

    • Adipocyte number is generally considered stable in mature humans and rodents.
    • Existing methods for measuring adipocyte proliferation do not differentiate between cell filling and new cell synthesis.

    Purpose of the Study:

    • To investigate the developmental timeline of adipocyte synthesis in Sprague-Dawley rats.
    • To determine if new adipocyte formation occurs postnatally and identify the timing of its cessation.

    Main Methods:

    • In vivo injection of [(3)H]thymidine in Sprague-Dawley rats.
    • Histometric analysis and Coulter counting of osmium-fixed cells were used to assess adipocyte populations.

    Main Results:

    • New adipocyte synthesis was observed to continue after birth in rats.
    • Adipocyte proliferation ceased before the onset of sexual maturity.
    • A significant "bed" of preadipocytes was synthesized during the second and third postnatal weeks.
    • The differentiation of preadipocytes into mature adipocytes can take up to 30 days.

    Conclusions:

    • Adipocyte development is a dynamic process during early postnatal life in rats.
    • Preadipocyte proliferation and differentiation are key mechanisms for adipose tissue expansion in early development.
