    Summary

    Inactivated target cells, not T lymphocytes, recognize responding cells in mixed lymphocyte reactions. This recognition, rather than T cell activity, explains observed immune responses to foreign cells.

    Background:

    • Mixed lymphocyte reactions (MLRs) are a key model for studying T cell-mediated immune responses.
    • Previous studies suggested T lymphocytes might not be required for MLRs, and F(1) hybrid lymphocytes could recognize parental alloantigens.

    Purpose of the Study:

    • To investigate the mechanism underlying mixed lymphocyte reactions.
    • To determine the role of T lymphocytes versus target cells in MLRs.
    • To re-evaluate the recognition of parental alloantigens by F(1) hybrid lymphocytes.

    Main Methods:

    • Mouse spleen cell populations depleted of thymus-derived (T) lymphocytes were cultured with inactivated allogeneic target cells.
    • F(1) hybrid responder cells were cultured with inactivated parental target cells.
    • Analysis of cellular interactions and recognition pathways in MLRs.

    • Observed MLRs occurred even when T lymphocytes were depleted.
