Abstract:
Thin layer chromatography as extraction method for histological sections. A combined microanalytical and histochemical procedure. A combined histochemical and chromatographical procedure is described to identify tissue components such as lipids, free amino acids, carbohydrates and steroids in cryosections. Fixed or unfixed sections are extracted by solvents of decreasing or increasing polarity. The extracts are then analysed by chromatographical techniques known from analysis of tissue homogenates. The extracted sections can be controlled by histochemical staining. Our technique is demonstrated in detail by analysis of lipids in bovine seminal vesicle epithelium, by analysis of free-amino acids in rat brain and pituitary and by analysis of steroids in male rat adrenal gland.
