A central vasodepressor effect of Dyflos.

H Edery, P G Guertzenstein

    British Journal of Pharmacology
    |April 1, 1974
    PubMed
    Summary

Dyflos application to the medulla oblongata caused vasodepression in cats. Acetylcholinesterase reactivators, obidoxime and pralidoxime, reversed this effect, suggesting a central mechanism.

    Area of Science:

    • Neuropharmacology
    • Cardiovascular Physiology

    Background:

    • Organophosphate compounds like dyflos can affect cardiovascular regulation.
    • The ventral medulla oblongata plays a critical role in controlling blood pressure and respiration.

    Purpose of the Study:

    • To investigate the effects of dyflos on cardiovascular parameters when applied to the ventral medulla oblongata in cats.
    • To determine if acetylcholinesterase reactivators can reverse dyflos-induced cardiovascular changes.

    Main Methods:

    • Dyflos was topically applied to the ventral medulla oblongata of anesthetized cats.
    • Cardiovascular parameters (arterial blood pressure, heart rate) and respiration were monitored.
    • Acetylcholinesterase reactivators (obidoxime, pralidoxime) and other agents were applied to assess their effects.

    Main Results:

    • Topical dyflos application induced vasodepression and altered respiration.
    • Atropine methyl nitrate prevented dyflos-induced vasodepression.
    • Obidoxime and pralidoxime mesylate reversed dyflos- and carbachol-induced vasodepression but not glycine-induced effects.

    Conclusions:

    • Obidoxime and pralidoxime may exert their effects through central acetylcholinesterase reactivation or a central atropine-like action.
    • The findings suggest a role for central cholinergic mechanisms in mediating the cardiovascular effects of dyflos.
