Blood volume determination in the mouse.

A C Riches, J G Sharp, D B Thomas

    The Journal of Physiology
    |January 1, 1973
    PubMed
    Summary

Accurate mouse blood volume measurement requires distinct red cell and plasma volume determinations using radiolabeled tracers. Relying on venous hematocrit can lead to significant discrepancies in blood volume calculations.

    Area of Science:

    • Physiology
    • Hematology
    • Animal Models

    Background:

    • Accurate determination of blood volume is crucial for physiological research.
    • Previous methods may have inaccuracies due to hematocrit variations.

    Purpose of the Study:

    • To accurately measure mouse blood volume using radiolabeled tracers.
    • To highlight the discrepancy between venous and whole-body hematocrit.

    Main Methods:

    • Utilized 59Fe-labeled red blood cells to measure red cell volume.
    • Employed 131I-labeled human serum albumin to determine plasma volume.
    • Calculated total blood volume from red cell and plasma volumes.

    Main Results:

    • Determined blood volumes for CSI female, CBA female, and CBA male mice (95.0 ± 1.5, 96.3 ± 2.7, and 84.7 ± 1.2 ml/kg body weight, respectively).
    • Observed a significant difference between venous (cardiac) hematocrit and whole-body hematocrit.

    Conclusions:

    • Mouse blood volume should be calculated using independently measured red cell and plasma volumes.
    • Venous hematocrit is not a reliable indicator for whole-body blood volume estimation in mice.
