    Background:

    • Platelets contain glycogen particles within their cytoplasm.

    Purpose of the Study:

    • To investigate the distribution of glycogen particles in rat platelets.
    • To examine the association of glycogen with the platelet open channel system.
    • To determine the stability of glycogen in relation to platelet activation and storage.

    Main Methods:

    • Electron microscopy was used to visualize glycogen distribution in rat platelets.
    • Platelets were subjected to various stimuli (thrombin, ADP, collagen) and storage conditions (21°C, EDTA, chilling).
    • The location and density of glycogen particles were analyzed in relation to the open channel system and granules.

    Main Results:

    • Glycogen particles were found both dispersed in the hyaloplasm and in masses near the open channel system in rat platelets.
    • Approximately one-third of platelets showed glycogen associated with the open channel system, often membrane-bound and near granules.
    • Stimulation and storage led to a loss of glycogen near the open channel system, while dispersed glycogen remained more stable.

    • Rat platelets exhibit a distinct localization of glycogen adjacent to the open channel system.
    • This open channel system-related glycogen appears more susceptible to degradation during platelet activation and storage.
    • The findings suggest a potentially specialized role for this labile glycogen pool compared to dispersed platelet glycogen.
