    Summary

    Fletcher factor deficiency impairs blood clotting and kinin generation. This study demonstrates that Fletcher factor is identical to prekallikrein, a key protein in these processes.

    Background:

    • Fletcher factor deficiency is associated with abnormal thromboplastin formation and kinin generation.
    • The precise identity and function of Fletcher factor in coagulation and kinin pathways were not fully elucidated.

    Purpose of the Study:

    • To investigate the role of Fletcher factor in plasma thromboplastin formation, kinin generation, and plasmin formation.
    • To determine if Fletcher factor is identical to prekallikrein.

    Main Methods:

    • Assessed thromboplastin formation in Fletcher factor-deficient plasma corrected with normal plasma or purified prekallikrein.
    • Measured kinin generation upon kaolin activation in deficient plasma with and without prekallikrein.
    • Evaluated PF/dil generation and plasmin formation in Fletcher factor-deficient plasma under various conditions.

    Main Results:

    • Fletcher factor-deficient plasma showed corrected thromboplastin formation and kinin generation upon addition of prekallikrein.
