Summary

Background:

Insulin is a key hormone regulating blood glucose.
Certain substances with higher molecular weights than insulin can react with anti-insulin antibodies.
These substances have been investigated in various biological samples and systems.

Purpose of the Study:

To describe and categorize substances with immunoreactivity to anti-insulin antibodies but higher molecular weights than insulin.
To review the discovery and characterization of these higher molecular weight forms.

Main Methods:

Analysis of pancreatic adenomas and extracts.
Study of serum samples.
Investigation in cell-free systems.
Use of anti-insulin antibodies for detection.

Main Results:

Proinsulin was identified as the first such substance.
Pre-proinsulin, the precursor to proinsulin, was subsequently discovered.
A group of substances with ill-defined composition, termed "high molecular weight immunoreactive insulin" (HWIRI), were also characterized.

Conclusions:

Multiple forms of insulin-related substances with higher molecular weights exist.
These HWIRI substances represent important components in understanding insulin biology and related pathologies.
