Site of action of nitrofurantoin in experimental urinary tract infection
Abstract:
The effectiveness of nitrofurantoin in suppressing bacterial growth in the urinary tract was evaluated by using two different experimental models. Pyelonephritis was produced in rats by direct inoculation of 10(4)Escherichia coli in the medulla of left kidney. Ascending urinary tract infection was induced by inoculation into the urinary bladder of 10(7)Proteus mirabilis, after a partial cystectomy. Nitrofurantoin was shown to be effective in suppressing bladder bacteriuria, in preventing ascending pyelonephritis, and also in preventing bacterial multiplication in kidney tissue following direct inoculation.
Insights
Nitrofurantoin effectively suppresses bacterial growth in the urinary tract. This antibiotic prevents bladder infections and kidney infections in rat models, demonstrating its therapeutic potential for urinary tract infections.
Area of Science:
- Pharmacology
- Microbiology
- Urology
Background:
- Urinary tract infections (UTIs) are common bacterial infections.
- Bacterial resistance and infection spread to the kidneys are clinical concerns.
- Nitrofurantoin is a widely used antibiotic for UTIs.
Purpose of the Study:
- To evaluate the efficacy of nitrofurantoin in suppressing bacterial growth within the urinary tract.
- To assess nitrofurantoin's effectiveness in preventing both lower and upper UTIs in experimental models.
Main Methods:
- Two distinct experimental models were employed to simulate urinary tract infections in rats.
- Pyelonephritis was induced by direct Escherichia coli inoculation into the kidney medulla.
- Ascending UTI was established via Proteus mirabilis inoculation into the bladder post-partial cystectomy.
Main Results:
- Nitrofurantoin demonstrated significant effectiveness in suppressing bacteriuria in the bladder.
- The antibiotic successfully prevented the development of ascending pyelonephritis.
- Nitrofurantoin also inhibited bacterial multiplication within kidney tissue following direct inoculation.
Conclusions:
- Nitrofurantoin exhibits potent antibacterial activity against common UTI pathogens in vivo.
- The drug is effective in preventing and treating bacterial infections at both bladder and kidney levels.
- These findings support the continued use of nitrofurantoin for managing urinary tract infections.
