Intrarenal complement fixation by cytotxic antibodies.

    Summary

This study shows that antibodies and complement proteins deposit in blood vessels during allotransplantation rejection. This pattern differs from glomerulonephritis and may aid in diagnosing graft rejection.

    Area of Science:

    • Immunology
    • Transplantation Biology
    • Pathology

    Background:

    • Histocompatibility antigens play a crucial role in transplant rejection.
    • Antibody and complement activation are implicated in graft damage.

    Purpose of the Study:

    • To investigate the localization of antibodies and complement in a short-term rat kidney allotransplantation model.
    • To differentiate the immune deposition pattern in graft rejection from that of glomerulonephritis.

    Main Methods:

    • Perfusion of Lewis and DA rat kidneys with cytotoxic antisera.
    • Analysis of tissue sections for fluorescent antibody and complement (beta(1C) protein) adsorption.

    Main Results:

    • Both cytotoxic antibodies and complement localized to the endothelial lining and arterial/venous walls.
    • The immune deposition pattern was distinct from that seen in glomerulonephritis.
    • Endothelial cell enlargement and nuclear protrusion were observed at deposit sites.

    Conclusions:

    • Antibody and complement deposition in graft vasculature suggests a role in rejection.
    • The distinct deposition pattern may help differentiate graft rejection from glomerulonephritis.
    • Findings support the hypothesis of antibody-mediated complement activation in allograft destruction.
