    Background:

    • Histamine is a crucial biogenic amine involved in various physiological processes.

    Purpose of the Study:

    • To investigate the primary pathways of endogenous histamine catabolism in mice.
    • To evaluate the efficacy of specific enzyme inhibitors on histamine inactivation.

    Main Methods:

    • Utilized radiolabeled (14)C-L-histidine to trace histamine formation.
    • Administered inhibitors of diamine oxidase (aminoguanidine), monoamine oxidase (pargyline), and histamine N-methyltransferase (methylhistamine).
    • Measured (14)C-histamine and (14)C-methylhistamine levels in mouse tissues and urine under various experimental conditions, including Freund's adjuvant pretreatment.

    • Diamine oxidase plays a minor role in the overall inactivation of newly formed histamine.
    • Measuring (14)C-histamine in tissues after aminoguanidine and methylhistamine administration is the preferred method for studying histamine formation.
    • Further development of specific methylhistamine assays and methylation inhibitors is warranted.
