A calcium pump in vascular smooth muscle.

D F Fitzpatrick, E J Landon, G Debbas

    Science (New York, N.Y.)
    |April 21, 1972

    Summary

Rabbit aorta microsomes, with magnesium and ATP, absorb calcium. This calcium uptake is slower than in skeletal muscle and limited in aorta mitochondria, which are also inhibited by azide.

    Area of Science:

    • Vascular biology
    • Cellular physiology
    • Biochemistry

    Background:

    • The intimal-medial layer of the aorta is crucial for vascular health.
    • Calcium homeostasis plays a vital role in cellular functions, including muscle contraction and signaling.
    • Understanding calcium handling in vascular cells is important for cardiovascular research.

    Purpose of the Study:

    • To investigate calcium uptake mechanisms in rabbit aortic microsomes and mitochondria.
    • To compare calcium uptake rates between aortic and skeletal muscle microsomes.
    • To determine the influence of specific ions and inhibitors on aortic calcium transport.

    Main Methods:

    • Preparation of microsomal and mitochondrial fractions from rabbit aorta.
    • Measurement of calcium uptake in the presence of magnesium and adenosine triphosphate.
    • Comparison of uptake rates with skeletal muscle microsomes.
    • Assessment of azide's effect on mitochondrial calcium uptake.

    Main Results:

    • Rabbit aortic microsomes actively transport calcium when supplied with magnesium and adenosine triphosphate.
    • The rate of calcium uptake by aortic microsomes is significantly slower compared to skeletal muscle microsomes.
    • Mitochondrial calcium uptake from the aorta is limited.
    • Azide, an inhibitor, effectively blocks calcium uptake by aortic mitochondria, but not by microsomes.

    Conclusions:

    • Aortic microsomes possess an active calcium uptake system, albeit with lower efficiency than skeletal muscle.
    • Aortic mitochondria have a restricted capacity for calcium uptake, sensitive to azide inhibition.
    • These findings contribute to understanding calcium regulation in vascular smooth muscle cells.
