    Summary

    N-methylhistamine and N-dimethylhistamine are more potent stimulators of gastric acid secretion than histamine in dogs and cats. Direct arterial delivery significantly enhances their stimulatory effects on parietal cells.

    Background:

    • Histamine is a known stimulator of gastric acid secretion.
    • Previous studies suggested methylated histamines might be more potent stimulators.

    Purpose of the Study:

    • To confirm and quantify the potency of N-methylhistamine and N-dimethylhistamine as gastric acid secretagogues compared to histamine.
    • To investigate the effect of administration route on the efficacy of these compounds.

    Main Methods:

    • Experiments were conducted on dogs and cats with Heidenhain pouches or gastric fistulas.
    • Various doses of histamine, N-methylhistamine, and N-dimethylhistamine were administered via systemic or arterial routes.
    • Gastric acid (HCl) output was measured to assess secretory response.

    Main Results:

    • N-methylhistamine and N-dimethylhistamine demonstrated greater potency than histamine in stimulating gastric acid secretion.
    • Arterial administration of methylated histamines resulted in significantly higher HCl output (4.1-13.9 times) compared to histamine.
    • N-dimethylhistamine showed twofold greater potency than histamine at the one-half maximal response level.

    Conclusions:

    • N-methylhistamine and N-dimethylhistamine are more potent stimulators of gastric acid secretion than histamine in dogs and cats.
    • Direct arterial delivery to the gastric mucosa enhances the stimulatory effect.
    • These findings suggest N-methylhistamine and/or N-dimethylhistamine may function as natural chemostimulators of parietal cells.
