    Background:

    • Antibiotic efficacy in treating prostatitis and urinary tract infections (UTIs) depends on drug penetration into target tissues and achieving synergistic concentrations.
    • Co-trimazine, a combination of trimethoprim and sulphadiazine, is used for bacterial infections.

    Purpose of the Study:

    • To determine the tissue and fluid concentrations of trimethoprim and sulphadiazine from co-trimazine in dogs.
    • To compare these concentrations with those of co-trimoxazole (trimethoprim and sulphamethoxazole).
    • To evaluate the pharmacokinetic profiles relevant to synergistic antimicrobial activity.

    Main Methods:

    • Constant infusion experiments were conducted in dogs.
    • Concentrations of trimethoprim and sulphadiazine were measured in serum, prostatic secretion, prostatic interstitial fluid, prostatic tissue, and urine.

    Main Results:

    • Trimethoprim concentrations were higher in prostatic tissues and fluids than in serum.
    • Sulphadiazine concentrations were significantly lower in prostatic tissues and fluids compared to serum.
    • Co-trimazine exhibited a trimethoprim/sulphadiazine ratio close to the optimal 1:20 synergy ratio in serum and urine, unlike co-trimoxazole.
    • Urine concentrations of sulphadiazine were substantially higher than those of sulphamethoxazole.

    Conclusions:

    • Neither co-trimazine nor co-trimoxazole shows a clear theoretical advantage for treating prostatitis based on these canine models.
    • Co-trimazine's high sulphadiazine urine concentrations suggest it may be more effective for general urinary tract infections.
