    Summary

    Background:

    • Ascaris suum is a common parasite causing allergic reactions.
    • Understanding the physiological mechanisms of allergic airway responses is crucial for asthma research.

    Purpose of the Study:

    • To investigate the physiological changes in conscious sheep following inhalation challenge with Ascaris suum extract.
    • To characterize the anaphylactic airway response and its relation to histamine release.

    Main Methods:

    • 10 conscious sheep with Ascaris suum sensitivity were challenged with Ascaris suum extract.
    • Measurements included pulmonary resistance (RL), lung compliance (Cst), functional residual capacity (FRC), arterial blood gases, and plasma histamine (H).
    • Control challenge with ragweed extract was performed.

    Main Results:

    • Five sheep developed significant bronchospasm (RL increase to 248%) and pulmonary hyperinflation (FRC increase to 126%) post-challenge.
    • Arterial oxygen (PO2) decreased significantly (to 63% of baseline).
    • Plasma histamine (H) increased markedly (to 423% of baseline) within 5 minutes, returning to baseline shortly after.

    Conclusions:

    • Antigen-induced bronchospasm in sensitized sheep is an anaphylactic airway response.
    • This response shares features with human bronchial asthma, including pulmonary hyperinflation.
    • Histamine plays a key role in mediating these acute allergic airway reactions.
