    Summary

    Intracytoplasmic lumina, or intracellular lumens, are frequently observed in various urothelial carcinomas in humans and canines. These structures, primarily within the cytoplasm, may play a role in tumor secretion processes.

    Background:

    • Intracytoplasmic lumina (ICLs) are intracellular structures observed in neoplastic cells.
    • Their presence and significance in urothelial carcinomas are not fully understood.

    Purpose of the Study:

    • To investigate the occurrence and characteristics of ICLs in spontaneous and chemically induced urothelial carcinomas.
    • To analyze the ultrastructural and histochemical features of ICLs.

    Main Methods:

    • Histochemical staining (PAS, alcian blue-PAS, mucicarmine) of paraffin and epoxy embedded sections.
    • Ultrastructural analysis using electron microscopy.
    • Examination of spontaneous and induced bladder carcinomas in humans, canines, and rats, including cell lines.

    Main Results:

    • ICLs were identified in spontaneous and induced urothelial carcinomas and derived cell lines.
    • Histochemically, ICLs stained positive for PAS and alcian blue-PAS, with uneven mucicarmine staining.
    • Ultrastructurally, ICLs were membrane-bound with microvilli and cytoskeletal elements; continuity with extracellular space was rare.

    Conclusions:

    • ICLs appear to be a relatively common finding in various urothelial carcinomas.
    • ICLs are predominantly, but not exclusively, intracytoplasmic.
    • The mechanisms of ICL formation and their relationship to secretion require further investigation.
