    Summary

    Background:

    • The official first action method (42.C01-42.C04) for determining carbadox in swine feed required modifications.
    • Issues with overheating and evaporation during sample preparation were noted with the original method.

    Purpose of the Study:

    • To modify and evaluate an improved analytical method for carbadox in swine feed.
    • To assess the performance of the modified method through a collaborative study.

    Main Methods:

    • The carbadox analysis method was modified by changing sample leaching from boiling for 1 hour to overnight leaching at room temperature.
    • The dilution scheme for spiked samples was adjusted to optimize spectrophotometer absorbance readings.
    • A collaborative study involving 21 laboratories was conducted to validate the modified procedure.

    Main Results:

    • Repeatability and reproducibility standard deviations were calculated for two different carbadox concentrations.
    • Statistical results, including mean percent of intent values (102% and 104%), were comparable to the official method.
    • A significant between-laboratory variance ratio was observed for feeds containing 0.0198% carbadox.

    Conclusions:

    • The modified carbadox analysis method demonstrated comparable statistical performance to the official first action method.
    • Despite modifications, the revised procedure is not recommended as a replacement for the current official method.
