    Summary

Solid-state acetylation of codeine phosphate with aspirin produces acetylcodeine phosphate. Gas-liquid chromatography (GLC) methods were developed for analyzing these compounds and their byproducts.

    • Pharmaceutical Chemistry
    • Analytical Chemistry
    • Chemical Kinetics

    Background:

    • Codeine phosphate is a widely used analgesic.
    • Understanding its solid-state reactions is crucial for drug formulation and stability.
    • Acetylation is a common modification for altering drug properties.

    Purpose of the Study:

    • To report a novel nonsolvolytic (solid-state) acetylation of codeine phosphate.
    • To develop and describe Gas-Liquid Chromatography (GLC) assays for simultaneous analysis of reactants and products.
    • To investigate the reaction kinetics and mechanisms involved.

    Main Methods:

    • Solid-state reaction between codeine phosphate and aspirin.
    • Development of GLC methods for quantifying aspirin, salicylic acid, codeine, and acetylcodeine.
    • Estimation of the apparent heat of activation for codeine phosphate.

    Main Results:

    • Successful synthesis of acetylcodeine phosphate via solid-state acetylation.
    • Validated GLC assays enable simultaneous determination of key compounds.
    • The apparent heat of activation for codeine phosphate was estimated, providing kinetic insights.

    Conclusions:

    • Solid-state acetylation offers a viable route for modifying codeine phosphate.
    • The developed GLC methods are effective for pharmaceutical analysis.
    • Kinetic data contributes to understanding the reaction mechanism and optimizing conditions.
