    Background:

    • Streptococcal preparation OK-432 (Picibanil) is a clinically used immunopotentiator.
    • OK-432 is known to activate the complement system in vitro via classical or alternative pathways.

    Purpose of the Study:

    • To investigate the effect of OK-432 on serum complement levels in vivo.
    • To identify specific complement components affected by OK-432 administration.

    Main Methods:

    • Hemolytic assays were performed using sensitized sheep erythrocytes (EA) for classical pathway and unsensitized rabbit erythrocytes (RaE) for alternative pathway.
    • Serum complement levels and specific complement components (C3, C1q, C4, C5, C9, properdin, C3 activator, C1-inhibitor) were analyzed.

    Main Results:

    • OK-432 increased overall serum complement levels in guinea pigs and humans without malignancy.
