    Background:

    • Kininogenase (KGA) and kininase activities are present in rat kidneys.

    Purpose of the Study:

    • To characterize and compare kininogenase (KGA) activity in rat kidneys and urine.
    • To investigate the enzymatic properties and relative abundance of KGA in renal and urinary compartments.

    Main Methods:

    • Gel filtration chromatography (Sephadex G-100) was used to separate kidney KGA and kininase activities.
    • Enzyme properties (molecular size, pH optimum, inhibitor effects) were compared between kidney and urinary KGA.
    • Hydrolysis of N-alpha-benzoyl-L-arginine ethyl ester (BAEE) was used to assess KGA activity.

    Main Results:

    • Kidney KGA activity was retained on Sephadex G-100, while kininase activity was excluded.
    • Kidney KGA exhibited identical molecular dimensions, pH optimum, inhibitor sensitivity, and kinin-releasing ability compared to urinary KGA.
    • 24-hour urine samples contained approximately 8 times more KGA activity than corresponding kidneys, with a low urine:kidney alkaline phosphatase ratio.

    Conclusions:

    • Rat kidneys contain distinct kininogenase (KGA) and kininase activities, with KGA primarily in sedimentable fractions.
    • Renal KGA is biochemically similar to urinary KGA, suggesting a common origin or shared characteristics.
    • Urinary KGA is significantly more abundant than renal KGA, indicating substantial renal excretion or production.
