    Summary

Guancydine effectively lowers blood pressure in hypertensive patients for up to 7 hours. This antihypertensive drug also reduces platelet aggregation and activates fibrinolysis, potentially preventing atherosclerosis and thrombosis.

    Area of Science:

    • Pharmacology
    • Cardiovascular Medicine

    Background:

    • Arterial hypertension is a significant risk factor for atherosclerosis and thrombosis.
    • Novel therapeutic agents are needed to manage hypertension and its complications.

    Purpose of the Study:

    • To evaluate the antihypertensive effects and potential prophylactic role of Guancydine.
    • To investigate the impact of Guancydine on angiotensin II response and related physiological parameters.

    Main Methods:

    • Clinical study involving nine hypertensive patients.
    • Administration of single oral doses of Guancydine (500-750 mg).
    • Monitoring of blood pressure, response to angiotensin II, and urinary excretion of electrolytes and water.

    Main Results:

  • Guancydine lowered blood pressure in 8 out of 9 patients for approximately 6-7 hours.
  • No impairment of vasopressor response to angiotensin II was observed.
  • Reduced excretion of water, Na, K, and Ca; decreased platelet adhesiveness; and activated fibrinolysis were noted.
  • Increased venous blood oxygenation and mild side effects like headache and orthostatic hypotension were reported.
  • Conclusions:

    • Guancydine demonstrates significant antihypertensive effects.
    • Its influence on platelet aggregation and fibrinolysis suggests a potential role in preventing cardiovascular complications.
    • Guancydine appears to be a safe antihypertensive agent with potential prophylactic benefits.
