    Summary

    Radiation exposure in mice can cause generalized lymphosarcomatosis. Viruses observed in these tumors did not appear to cause cancer, suggesting they are not responsible for the disease development.

    Background:

    • Generalized lymphosarcomatosis, a type of non-thymic leukemia, is observed in mice exposed to radioactive isotopes like strontium-90 (90Sr), plutonium-239 (239Pu), or radium-226 (226Ra).
    • The presence of viruses within these radiation-induced tumors raises questions about their etiological role, particularly if they are activated by irradiation.

    Purpose of the Study:

    • To investigate whether tumors arising from radiation exposure in mice possess tumor-associated transplantation antigens.
    • To determine if these potential antigens, if virus-determined and activated by irradiation, could elicit a protective immune response.

    Main Methods:

    • Passaging of tumors from irradiated mice bearing generalized lymphosarcomatosis.
    • Testing for protective immunity using different doses and types of tumor inoculations: sub-threshold doses of living syngeneic tumor, large doses of living allogeneic tumor, and large doses of killed syngeneic tumor.
    • Electron microscopy to observe viral presence in the tumors.

    Main Results:

    • No protective immunity was observed following challenges with sub-threshold doses of living syngeneic tumor.
    • Large doses of living allogeneic tumor did not induce protective immunity.
    • Large doses of killed syngeneic tumor also failed to confer protection.

    Conclusions:

    • The lack of protective immunity suggests that any tumor-associated transplantation antigens are not determined by viruses activated by irradiation.
    • Viruses observed via electron microscopy in these radiation-induced lymphosarcomas are likely passengers and not the causative agents of the disease.
