    Background:

    • Alpha2-Macroglobulin (alpha2M) is a major plasma protease inhibitor.
    • The role of alpha2M on the surface of endothelial cells is not well understood.

    Purpose of the Study:

    • To investigate the presence and localization of alpha2-Macroglobulin (alpha2M) on the luminal surface of human endothelial cells.
    • To determine if alpha2M on endothelial cells has a protective function against proteases.

    Main Methods:

    • Indirect immunofluorescence technique was used to detect alpha2M on human artery, vein, and lymphatic endothelial cells.
    • Immunoabsorption studies were performed to confirm the specificity of the immunofluorescent staining.
    • Control staining with antisera against other proteins (alpha1-antitrypsin, antithrombin III, IgG, IgA, IgM, C3, fibrinogen) was conducted.

    Main Results:

    • Alpha2-Macroglobulin (alpha2M) was identified on the luminal surface of endothelial cells in normal human arteries, veins, and lymphatics.
    • Specificity of the staining was confirmed by immunoabsorption, which completely inhibited staining.
