    Summary

    Apomorphine effectively stimulates growth hormone release in normal subjects, outperforming levodopa. This study compared apomorphine and levodopa for growth hormone stimulation, finding apomorphine more potent with manageable side effects.

    Background:

    • Growth hormone (GH) release is pulsatile and influenced by various stimuli.
    • Dopaminergic pathways play a role in GH regulation.
    • Levodopa is a known stimulant of GH release, but its efficacy can be variable.

    Purpose of the Study:

    • To compare the efficacy of apomorphine and levodopa in stimulating growth hormone (GH) release in normal volunteers.
    • To evaluate the safety and side effect profile of subemetic doses of apomorphine for GH stimulation.

    Main Methods:

    • Ten healthy volunteers (5 male, 5 female) participated.
    • Subemetic doses of apomorphine (0.76 mg subcutaneously) and levodopa (500 mg orally) were administered.
    • Serum growth hormone levels were measured to assess the response.

    Main Results:

    • Apomorphine induced a normal or borderline normal GH response in all 10 subjects.
    • Levodopa failed to elicit a normal GH response in 4 subjects and produced borderline or normal responses in the remaining 6.
    • Peak GH response was significantly higher with apomorphine (26.94 ng/ml) compared to levodopa (9.76 ng/ml) (p < 0.025).
    • Side effects included nausea and vomiting with both drugs, and transient drowsiness with apomorphine.

    Conclusions:

    • Apomorphine is a potent stimulant of GH release in normal individuals, demonstrating superior efficacy compared to levodopa.
    • Subemetic doses of apomorphine appear to be a reliable method for GH stimulation testing.
    • Further research may explore apomorphine's role in conditions with impaired GH secretion.
