Channels in planar bilayers made from commercially available lipids.

H Gögelein, H Koepsell

Pflugers Archiv : European Journal of Physiology
    |August 1, 1984
    PubMed
    Summary

Spontaneously fluctuating channels in planar bilayer membranes were observed using lipids from biological sources. These channels were absent in synthetic lipid bilayers, suggesting they originate from proteolipid impurities.

    Area of Science:

    • Biophysics
    • Membrane Science
    • Lipid Bilayer Research

    Background:

    • Planar bilayer membranes are crucial models for studying cell membrane properties.
    • Understanding ion channel behavior in lipid bilayers is essential for various biological processes.
    • Commercially available lipids are frequently used in creating these model membranes.

    Purpose of the Study:

    • To investigate the origin of spontaneously fluctuating channels observed in planar lipid bilayer membranes.
    • To determine whether lipid source (biological vs. synthetic) influences channel activity.
    • To identify the components responsible for observed channel activity.

    Main Methods:

    • Preparation of planar bilayer membranes using commercially available lipids, including those from biological sources (egg yolk phosphatidylcholine, soybean phosphatidylcholine, bovine brain phosphatidylserine) and synthetic sources (1,2 diphytanoyl 3-sn-phosphatidylcholine).
    • Observation and characterization of spontaneously fluctuating channel activity within the prepared lipid bilayers.
    • Comparative analysis of channel activity between membranes formed from biological and synthetic lipids.

    Main Results:

    • Spontaneously fluctuating channels were observed in planar bilayers composed of lipids derived from biological sources.
    • No spontaneous channel activity was detected in planar bilayers formed using synthetic lipids.
    • The presence of specific lipid types from biological origins correlated with the observation of channel activity.

    Conclusions:

    • The observed spontaneously fluctuating channels in planar lipid bilayers are attributed to proteolipid impurities present in the lipid material.
    • Lipids of biological origin are more likely to contain these proteinaceous impurities that form functional channels.
    • Synthetic lipids, being more purified, do not exhibit this channel activity, highlighting the importance of lipid purity in membrane studies.
