    Summary

    Human amniotic fluid contains angiotensin I-converting enzyme (ACE). This study found ACE and other components of the renin-angiotensin system in amniotic fluid, suggesting its presence during pregnancy.

    Background:

    • Angiotensin I-converting enzyme (ACE) plays a crucial role in the renin-angiotensin system.
    • The presence and function of ACE in human amniotic fluid are not well-established.

    Purpose of the Study:

    • To characterize the antigenic and enzymatic properties of ACE in human amniotic fluid.
    • To investigate the presence of other components of the renin-angiotensin system in amniotic fluid.

    Main Methods:

    • Radioimmunoassay was used to quantify ACE antigen levels.
    • Enzymatic activity was measured using Hip-His-Leu as a substrate.
    • Inhibition studies with captopril and antibodies were performed.

    Main Results:

    • ACE was detected in soluble form in human amniotic fluid (mean level 136 +/- 83 ng/ml).
    • Amniotic fluid ACE showed complete cross-reactivity with antibodies against lung ACE.
    • Significant correlation was found between ACE antigen levels and activity.
    • Angiotensinase, kininase, renin, and angiotensinogen were also identified in amniotic fluid.

    Conclusions:

    • A complete, though not fully activated, renin-angiotensin system exists in human amniotic fluid.
    • ACE in amniotic fluid shares antigenic and enzymatic properties with ACE from other human tissues.
    • These findings suggest a potential role for the renin-angiotensin system in the amniotic fluid environment during pregnancy.
