    Purpose of the Study:

    • To isolate and characterize circular DNA from ram spermatozoa mitochondria.
    • To compare the physicochemical properties of ram sperm mitochondrial DNA with its nuclear DNA.

    • Isolation of circular DNA from ram spermatozoa mitochondrial fractions using SDS treatment and sucrose gradient centrifugation.
    • Determination of DNA contour length, buoyant density, and melting temperature (Tm).

    • Circular mitochondrial DNA was successfully isolated.
    • Mitochondrial DNA exhibited a contour length of 5.0 micron.
    • Its buoyant density (1.6983 g cm-3) and Tm (69.7°C) were distinct from nuclear DNA components.

    • Ram spermatozoa contain distinct circular mitochondrial DNA.
    • Physicochemical properties of ram sperm mitochondrial DNA differ significantly from nuclear DNA.
    • This provides foundational data for understanding sperm mitochondrial genetics in rams.
