    Background:

    • Gastric acid and pepsin secretion are crucial for digestion but can cause issues when overproduced.
    • Histamine H2-receptor antagonists like ranitidine and cimetidine are used to reduce gastric secretion.

    Purpose of the Study:

    • To compare the inhibitory effects of ranitidine and cimetidine on gastric secretion.
    • To determine the relative potency and duration of action of these two H2-receptor antagonists.

    Main Methods:

    • Eight healthy volunteers participated in the study.
    • The effects of various doses of ranitidine and cimetidine on pentagastrin-stimulated gastric volume, acid, and pepsin secretion were measured.

    Main Results:

    • Both ranitidine and cimetidine demonstrated a dose-dependent inhibition of gastric volume, acid, and pepsin secretion.
    • The average inhibitory dose 50 (ID50) was approximately 5 mg for ranitidine and 41 mg for cimetidine.
    • Ranitidine was found to be approximately 11 times more potent than cimetidine on a molar basis, with similar durations of action.

    Conclusions:

    • Ranitidine exhibits significantly greater potency in inhibiting gastric acid and pepsin secretion compared to cimetidine.
    • Both drugs provide a comparable duration of action in suppressing gastric secretion.
