    Summary

The T25 molecule, which carries the Thy-1.2 antigen, shows structural variations in T lymphocytes. These differences in T25 structure correlate with T cell maturation and function.

    Area of Science:

    • Immunology
    • Molecular Biology
    • Cell Biology

    Background:

    • The Thy-1.2 antigen is a cell surface marker found on thymocytes and T lymphocytes.
    • The T25 molecule is associated with the Thy-1.2 antigen.
    • Understanding T25 structure is crucial for elucidating T cell development and function.

    Purpose of the Study:

    • To investigate the structural heterogeneity of the T25 molecule on different T cell populations.
    • To determine if T25 structural variations correlate with T lymphocyte maturation or function.

    Main Methods:

    • Isolation of T25 molecule from thymocytes and cloned T cells using immune precipitation with monoclonal antibodies.
    • Electrophoretic analysis of T25 polypeptides using SDS-PAGE.
    • Separation of thymocytes based on Thy-1.2 surface antigen density.

    Related Experiment Videos

    Main Results:

    • T25 molecules exhibited a heterogeneous pattern of polypeptide bands with apparent molecular weights ranging from 28,000-36,000 daltons.
    • Structural variations in T25 were observed between thymocyte subpopulations with different Thy-1.2 densities.
    • Differences in T25 band patterns were also detected in cloned cytolytic and helper T cells.
    • T25 structural heterogeneity varied between different cell types and populations.

    Conclusions:

    • Surface T25 molecules are structurally heterogeneous.
    • T25 structural variations are linked to T lymphocyte maturation and/or function.
    • These findings provide insights into the molecular basis of T cell differentiation and effector functions.
